San Jose, CA, United States., September 02, 2019 -- Neuropilins (NRPs) are multifunctional cell surface receptors that are expressed in all vertebrates and highly conserved across species. These proteins play a role in cellular processes such as development, axonal guidance, angiogenesis, cancer, immunity, cell motility, lymphangiogenesis, endothelial and tumor cell migration, and bone homeostasis. The two isoforms, NRP1 and NRP2 mainly act as co-receptors for Semaphorins and for members of the VEGF family of molecules. Neuropilin-1( NRP1) is a protein that in humans is encoded by the NRP1gene. NRP1 is expressed widely in adult tissues, including organs in which angiogenesis occurs such as heart and placenta. It has been implicated in the vascularization and progression of cancers.

NRP1 expression has been shown to be elevated in tumor samples, including brain, prostate, breast, colon, and lung cancers and NRP1 levels are positively correlated with metastasis. NRP1 was identified as a receptor for Collapsin-1/Semaphorin III/D (Sema III) and VEGF. Neuropilin 2 (NRP2) is a type I transmembrane glycoprotein that in humans is encoded by the NRP2 gene. It shows high affinity binding to VEGF and semaphorins. In addition to its role in neuronal development, NRP2 is important for the development of capillaries, lymphatic vessels and progression of cancers. It is expressed in various human cancer tissues and cancer cell lines. NRP2 axis regulates the trafficking of EGFR in cancer cells. Inhibitors of NRP2 should show a potent antitumor effect in cancer cells with activated EGFR signaling. VEGF-C/NRP-2 axis is involved in the activation of autophagy, which helps cancer cell survival following treatment. NRP2 in macrophages promotes tumor growth by regulating efferocytosis of apoptotic tumor cells and orchestrating immune suppression.WDFY1 is the downstream effector of NRP2. Regulation of WDFY1 transcription by NRP2 axis is a critical event in maintaining metastatic phenotype in cancer cells. Thus, inhibiting NRP2 can be an effective strategy to induce cell death in metastatic cancer.
