DEAE – Diethyl Amino Ethyl Cellulose columns are widely used for protein purification. DEAE cellulose is an anionic resin which can be widely used to bind negatively charged proteins. When a fractionation with ammonium sulphate precedes ion-exchange chromatography in a purification procedure often help to remove excess salt prior application of the protein mixture to the column. Basically it is done to ensure that the ionic strength of the solution is sufficiently low for absorption of at least some of the proteins in the mixture.
